Side EffectsMegaloblastic anaemia, pancytopenia, desquamation and bleeding may occur in GIT.
Skin reactions, alopecia, ocular irritation, arachnoiditis in intrathecal use, megaloblastic anaemia, osteoporosis, precipitation of diabetes, arthralgias.
Interstitial lung disease, leukoencephalopathy.
IndicationsMaintaining remission in children with acute leukaemias.
Autoimmune diseases like rapidly progressing rheumatoid arthritis, severe psoriasis, pemphigus, myasthenia gravis, uveitis, chronic active hepatitis.
Contra IndicationsPregnancy and lactation.
Severe renal or hepatic impairment, pre-existing profound bone-marrow suppression.
CautionEarly hepatic and renal dysfunction.
Bone marrow depression, elderly, neonates.
Use of folinic acid rescue has permitted much higher doses of methotrexate.
